The expression and prognostic value of GPNMB,p-AKTs473,p-GSK3βser9 and Ki-67 in esophageal squamous cell carcinoma
Objective To explore the expression of GPNMB,p-AKTs473,p-GSK3βser9 and Ki-67 and their relationship with clini-copathological prognosis in esophageal squamous cell carcinoma.Methods:The differentially expressed proteins were screened by ITRAQ,tissue chip and Immunohistochemical technique was used to detect the expression of GPNMB,p-AKTs473,p-GSK3βser9 and Ki-67 in all samples.The relationship between the above proteins and the clinicopathological features of the patients was analyzed by x 2 test.K-M survival analysis and COX regression were used to evaluate the effect of these proteins on the prognosis of patients with esophageal squamous cell carcinoma.Results:The positive expression rates of GPNMB,p-AKTs473,p-GSK3βser9 and Ki-67 in ESCC were higher than those in adjacent normal tissues.The positive expression of p-AKTs473 was associated with nerve invasion(P=0.03),and the positive expression of Ki-67 was associated with differentiation(P=0.006)and vascular invasion(P=0.000).The co-positive expression of the four proteins was correlated with lymph node metastasis(P=0.04)and vascular invasion(P=0.02).p-GSK3βser9 positive patients had shorter OS,Ki-67 positive patients had significantly shorter PFS,and the four proteins co-positive pa-tients had shorter OS and PFS.Conclusion:Compared with the positive expression of individual proteins,the co-expression of GPNMB,p-AKTs473,p-GSK3βser9 and Ki-67 may be related to the progression of ESCC.GPNMB is expected to be a potential target for optimizing therapeutic strategies for esophageal cancer.
