Effect of Rumex L.extract on the proliferation of human immortalized epidermal keratinocytes(HaCaT cells)
Objective Chemical components analysis of alcohol extracts from Rumex L.root and its effects on the proliferation of Human Immortalized Keratinocytes(HaCaT cells).Methods Using D101 Macroporous resin for separation purification in the Rumex L.70％ethanol extract,the main chemical components was screened by HPLC analysis,and Human Immortalized epidermal Keratino-cytes(HaCaT)proliferation was also observed.Results 30％,50％,70％and 90％ethanol elution parts were obtained by D101 macro-porous resin treatment and their extraction amounts were 5.4％,14.8％,11％and 0.72％,respectively.HPLC examine results showed that the 30％ethanol elution parts(30％EP)was 0.151％,and its components were emodin;the 50％ethanol elution parts(50％EP)was 1.952％,and its components were aloe emodin,rhein,emodin,and physcion;the 70％ethanol elution parts(70％)was 21.409％,and its components were aloe emodin,rhein,emodin and physcion;90％ethanol elution parts(90％EP)was 1.927％,and its components were emodin,rhein and physcion;the 70％ethanol extract(original crude extract)was 1.591％,and its components were aloe emodin and rhein.The inhibition HaCaT cells proliferation from strong to weak were 90％EP,70％EP,50％EP,70％RE(original crude ex-tract)and 30％EP,with an inhibition rate of 17.55％,16.66％,10.99％,10.25％and 7.86％,respectively.Conclusion 70％EP was the best concentration parts(21.409％)and also was the best elution parts.Rumex L.70％ethanol extract and different ethanol elution parts can inhibit of HaCaT cell proliferation.Its effect may be related to anthraquinones composition in Rumex L.,suggesting that Ru-mex L.has the effect of treating psoriasis.
