Study on the Effects of Phenylalanine and Methyl Jasmonate on Profile of Essential Oil from Tissue Culture of Thymus vulgaris L.
The effects of phenylalanine(Ple) and methyl jasmonate(MJ) on growth of in vitro adventitious buds of Thymus vulgaris L.and profile of their essential oils were studied.The results showed that by adding different concentrations of Ple and MJ,the proliferation of adventitious buds could be promoted,and the best proliferation was found when 100 mg/L Ple and 150 μmol/L MJ were added,compared with the control,the proliferation coefficients were significantly increased 47.03 % and 23.57% respectively.The essential oils from the tissue culture seedlings,and the tissue culture seedlings treated with phenylalanine(Ple) and methyl jasmonate(MJ)were extracted by steam distillation,the chemical constituents and their relative contents of these three samples were identified and analyzed by gas chromatography-mass spectrometry with peak area normalization method.The result showed that the main constituents of these three samples had no significant difference,they all mainly contained thymol,carvacrol,o-cymene,γ-terpinene,caryophyllene,etc.,only slight differences in the relative contents were observed.This meant suitable concentration of Ple and MJ could increase proliferation coefficient of adventitious buds of Thymus vulgaris L.and improve the physiological activities of the plant,also,the accumulation and synthesis of secondary metabolites within this palnt were promoted.
