摘要

探讨复方精油经鼻吸入对睡眠剥夺小鼠的清醒维持作用及机制.动物试验结果表明,复方精油不仅能显著提高睡眠剥夺小鼠自主活动(P<0.01),显著缩短睡眠时间(P<0.01),具有维持清醒的作用,还能使超氧化物歧化酶(SOD)和谷胱甘肽过氧化物酶(GSH-Px)活性及脑内多巴胺(DA)和谷氨酸(Glu)水平显著升高(P<0.05,P<0.01),且显著降低 5-羟色胺(5-HT)水平(P<0.01);细胞结果表明,复方精油组多巴胺转运蛋白(DAT)、五羟色胺 1A(5-HT1A)蛋白表达极显著降低(P<0.01),5-羟色胺转运蛋白(SERT)、囊泡谷氨酸转运蛋白 1(VGLUT1)蛋白表达显著提高(P<0.05,P<0.01).

Abstract

To investigate the effect and mechanism of nasal inhalation of compound essential oil on awake maintenance in sleep-deprived mice.The results of animal experiments showed that the compound essential oil could not only significantly improve the spontaneous activity of sleep deprivation mice(P<0.01),significantly shorten the sleep time(P<0.01),and maintain awake,but also significantly increase the activities of superoxide dismutase(SOD)and glutathione peroxidase(GSH-Px)and the levels of dopamine(DA)and glutamic acid(Glu)in the brain(P<0.05,P<0.01),and significantly reduce the level of 5-hydroxytryptamine(5-HT)(P<0.01).The cell results showed that the expression of dopamine transporter(DAT)and serotonin 1A(5-HT1A)protein in the compound essential oil group decreased significantly(P<0.01),and the expression of serotonin transporter(SERT)and vesicular glutamate transporter 1(VGLUT1)protein increased significantly(P<0.05,P<0.01).
