液相色谱-三重四级杆质谱法测定化妆品中51种抗组胺类药物的含量

建立了液相色谱-三重四级杆质谱法(LC-MS-MS)快速测定化妆品中地氯雷他定等51 种抗组胺类药物含量的方法.样品经 10 mmol/L乙酸铵甲醇溶液超声提取后,经 0.2 μm滤膜过滤后,以 10 mmol/L乙酸铵水-甲醇作为流动相进行梯度洗脱,经Eclipse Plus C18 色谱柱(100 mm×3.0 mm×1.8 μm)分离,采用电喷雾离子源在正、负离子模式下进行多反应监测,外标法定量.结果表明,51种抗组胺类药物在2~50 ng/mL范围内线性关系良好(r>0.999),检出限和定量限分别为 0.15 μg/g和 0.5 μg/g.对液态水基、乳液、膏霜、面膜、液态油基、凝胶、粉、蜡基 8 种不同化妆品基质在0.5、1.0、5.0 μg/g加标水平下的平均回收率为70.3%~127.8%,相对标准偏差(RSD)小于7.7%(n=6).该方法前处理操作简便、快速、专属性强、灵敏度高、精密度、准确度均较好,可用于化妆品中 51 种抗组胺类药物含量的快速测定.
Determination of 51 Antihistamines in Cosmetics by High Performance Liquid Chromatography-Tandem Mass Spectrometry
A high performance liquid chromatography-tandem mass spectrometry(LC-MS-MS)method was established for the simultaneous detection of 51 antihistamines including desloratadine in cosmetics.The samples were extracted ultrasonically with 10 mmol/L ammonium acetate methanol solution,and then filtered through 0.2 μm membrane.The analytes were separated by Eclipse Plus C18 column(100 mm×3.0 mm×1.8 μm)with a gradient elution of 10 mmol/L ammonium acetate and methanol.The analytes were quantified by the external standard method in multiple reaction monitoring(MRM)mode with electrospray ion source in positive and negative modes.The correlation coefficients of the standard calibration curves were greater than 0.999 in the range of 2-50 ng/mL.The limits of detection and limits of quantification were 0.15 µg/g and 0.5 µg/g,respectively.The average recoveries of 51 antihistamines ranged from 70.3%to 127.8%at the spiked levels of 0.5,1.0,5.0 µg/g in 8 different cosmetics matrices including water-based liquid,lotion,cream,face-mask,oil,gel,power and wax,with the relative standard deviations(RSDs)less than 7.7%(n=6).The developed method is simple,rapid,exclusive,sensitive,accurate and precise,and can be applied in the quantitative of 51 antihistamines in cosmetics.
