摘要

建立了一种使用高效液相色谱测定化妆品中羟基积雪草苷和积雪草苷含量的方法.样品经提取液超声提取后,用Acutfex PA-C18 色谱柱(250 mm×4.6 mm×5 μm)分离,以体积比2:2:6 的甲醇-乙腈-水作为流动相等度洗脱,流速1.0 mL/min,柱温35℃,检测波长 205 nm.采用外标法测定,结果表明,积雪草苷和羟基积雪草苷在质量浓度5~100 μg/mL内线性关系良好,方法的检出限为质量分数0.001 5%,定量限为质量分数0.005%.在不同类型的空白化妆品(水类、乳类、膏霜类)中进行加标回收试验,方法的回收率为 82.8%~106%,相对标准偏差为0.94%~4.28%.该方法前处理简单,分离效率高,重复性好,准确度和精密度高,适用于化妆品中积雪草苷和羟基积雪草苷的测定.

Abstract

A high performance liquid chromatography(HPLC)method for determination of madecassoside and asiaticoside was developed.The sample was extracted with extraction liquid by ultrasonic technique,and then separated on Acutfex PA-C18 column(250 mm×4.6 mm×5 μm),eluted with a mixture of methanol-acetnirile-water in the volume ratio of 2:2:6 as the isocratic mobile phase.The flow rate was 1.0 mL/min and the temperature of column was 35℃,and detected at a wavelength of 205 nm.The external standard method was used for quantitative determination.The results showed that the linear relationship of madecassoside and asiaticoside were good within the concentration range of 5-100 μg/mL,and the detection limit was 0.001 5%(in mass fraction),and the quantification limit was 0.005%(in mass fraction).The spiked recovery test was carried out in different types of cosmetics(water,milk,cream),and the recoveries of the method were 82.8%-106%with the relative standard deviations of 0.94%-4.28%.The method is simple in pretreatment,efficient in separation,reproducible,accurate and precise,and suitable for the determination of madecassoside and asiaticoside in cosmetics.
