摘要

采用分步提取的策略从淡黑雷松藻中制备一种低聚糖提取物(AIV1).AIV1 相对分子质量约为 8 000,糖醛酸相对含量为 63.64%,其单糖组成以甘露糖醛酸(ManA)和半乳糖(Gal)为主,并采用傅里叶-红外光谱和核磁共振波谱法对其结构进行表征.AIV1 对小鼠单核巨噬细胞(RAW264.7 细胞)和人皮肤成纤维细胞(HSF)无明显毒性,可显著降低脂多糖诱导的RAW264.7 细胞中一氧化氮的过量释放(P<0.01)、下调肿瘤坏死因子α(P<0.01)和白细胞介素 1β(P<0.05)mRNA的过量表达,同时可抑制长波紫外线(UVA)损伤造成的HSF细胞中活性氧的过量释放(P<0.05).因此,AIV1 具有良好的抗炎及抗光老化活性,可作为一种天然原料用于抗炎和抗光老化功效性化妆品的开发.

Abstract

Oligosaccharide extract(AIV1)was prepared from Lessonia nigrescens by step-by-step extraction strategy.AIV1 had a relative molecular mass of approximately 8 000,with a uronic acid relative content of 63.64%,and its monosaccharide composition was primarily composed of ManA and Gal.Fourier transform infrared spectroscopy(FTIR)and nuclear magnetic resonance(NMR)were utilized to characterize its structure.AIV1 had no significant toxicity to mouse monocyte-macrophages(RAW264.7)cells and human skin fibroblasts(HSF)cells.AIV1 could significantly reduce the excessive release of nitric oxide(P<0.01)in lipopolysaccharide-induced RAW264.7 cells,down-regulate the overexpression of tumor necrosis factor-α(TNF-α,P<0.01)and interleukin-1β(P<0.05)mRNA,and inhibit the excessive release of reactive oxygen species(ROS)in long wave ultraviolet(UVA)-damaged HSF cells(P<0.05).Therefore,AIV1 exhibited good anti-inflammatory and anti-photoaging activities,suggesting its potential as a natural ingredient for the development of anti-inflammatory and anti-photoaging functional cosmetics.
