摘要

为研究单次十二烷基硫酸钠(SLS)刺激和胶带剥离短效模型建模过程中的技术要点,采用不同质量分数的SLS溶液封闭刺激一侧前臂 24 h,撕下斑贴后的 1、2、4、6、24、48、72 h测试皮肤经皮失水流失(TEWL)和皮肤红斑指数(EI值);另一侧手臂取3个3 cm×3 cm区域,进行不同程度的胶带剥离,记录剥离前、剥离后的即刻(Tim)、2 h、4 h参数值.结果表明,对于单次SLS刺激模型,当SLS溶液质量分数为 1%时,24 h后,EI值达到最大值;当SLS溶液质量分数为 2%时,24 h后,TEWL与EI值均显著提升,且均达到最大值;对于胶带剥离模型,4 h后EI值反而明显升高.因此,并不是屏障损伤越严重短效模型效果越好,应该根据产品作用机理及测试目的对试验设计进行考量.

Abstract

The technical gist of modeling process in single sodium lauryl sulfate(SLS)stimulation and tape stripping short-term model was studied in this paper.By using SLS solutions with different mass fractions and stimulated one forearm under closed conditions for 24 h,skin transdermal water loss(TEWL)and skin erythema index(EI value)were measured at 1,2,4,6,24,48,72 h after the patch was removed.Take three 3 cm×3 cm areas from the other arm and perform different degrees of tape stripping.Record the parameter values of before stripping,immediate(Tim),2 h,4 h after stripping.The results showed that for a single SLS stimulation model,when the mass fraction of SLS solution was 1%,EI values reached the maximum value after 24 h.When the mass fraction of SLS solution was 2%,after 24 h,both TEWL and EI values reached their maximum value.For the tape stripping model,EI values increased significantly after 4 h.Therefore,it was not mean that the more severe the barrier damage,the better the short-term models.The experimental design should be considered based on the product's mechanism of action and the test purpose.
