壬二酸-壳聚糖凝胶体系的制备及其体外释放特性

采用壳聚糖、氨丁三醇和特定的多元醇溶剂体系,在pH 5.0~6.0条件下,成功制备了均一、透明、稳定的含壬二酸的凝胶体系.通过对壬二酸和壳聚糖质量比、多元醇种类及含量、pH范围、pH调节剂种类进行系统研究,使体系中壬二酸质量分数提高至 20%.选择质量分数 10%壬二酸对凝胶体系的结构进行表征和体外释放性能研究,红外光谱(IR)分析和热重-差示扫描量热(TG-DSC)联合分析结果显示壬二酸发生了成盐反应.体外释放性能测试结果显示该凝胶体中,壬二酸释放性能优异,释放速率达(3 193.61±75.45)μg/(cm2·min1/2),150 min累积释放率达(86.46%±3.04%),显著提高了壬二酸的生物利用度.
Preparation of Azelaic Acid-Chitosan Gel and Its Release Characteristics in vitro
Homogeneous,transparent and stable gel containing azelaic acid was successfully prepared by chitosan,tromethamine and specific polyol solvent system at pH 5.0-6.0.Through systematic research on the mass ratio of azelaic acid and chitosan,the type and content of polyols,pH range and the type of pH regulator,the content of azelaic acid in the system was increased to 20%(in mass fraction).The 10%azelaic acid gel was selected to characterize the structure and study its release performance in vitro.The combined analysis of infrared spectroscopy(IR)and thermogravimetric-differential scanning calorimetry(TG-DSC)showed that azelaic acid underwent a salt formation reaction.The in vitro release test results showed that azelaic acid in the gel had excellent release performance.The release rate was(3 193.61±75.45)μg/(cm2·min1/2)and the accumulated release rate was(86.46%±3.04%)after 150 min.The azelaic acid gel prepared in this study significantly improves the bioavailability of azelaic acid.
