Simultaneous Determination of 14 Antioxidant Active Components in Olea Europaea by HPLC
A high performance liquid chromatography(HPLC)method was established to determine the contents of 14 antioxidant ingredients(hydroxytysol,tyrosol,chlorogenic acid,vanilic acid,caffeic acid,ferinic acid,acteoside,luteolin,rutinose,oleuropein,quercetin,luteolin,kaempferol,apigenin)in Olea europaea.Samples were extracted by sonication with 70%(in volume fraction)methanol water solution,gradient eluted with methanol,0.1%(in volume fraction)phosphate acid solution as the mobile phase,separated by InfinityLab Poroshell 120 EC-C18(150 mm×4.6 mm×4 μm)column,and quantified by external standard.In the range of 1-100 mg/L,the mass concentration of 14 chemical components had a good linear relationship with their peak area(R2>0.999 8).The recovery rates were 95.3%-107.4%,the relative standard deviations(RSDs)were less than 1.6%(n=6).This method has the advantages of high precision and good repeatability,and can meet the requirements of quantitative detection of 14 compounds in Olea europaea,for raw material control and quality evaluation of Olea europaea.
