Percutaneous Absorption Process of Nicotinamide by in vitro Method
The percutaneous absorption of nicotinamide was investigated by diffusion cells method,Raman spectroscopy and fluorescence labelling method after different times of application on pig skin.The results of the diffusion cells method showed that the nicotinamide was able to enter into the receiving cell only after 24 h,and the subcutaneous diffusion rate was 21.98%.The results of Raman spectroscopy showed that niacinamide was absorbed at a depth of 30 μm after 1 h on the skin,and the percutaneous absorption was detected at a depth of 0~100 μm after 4 h and 24 h.The relative total amounts of percutaneous absorption were 0.250 3,0.656 3 and 1.751 7 a.u.,respectively.The results of the fluorescent labelling method showed that with the time increasing,the percutaneous absorption and its depth of nicotinamide were gradually increasing.The results showed that with the time increasing,niacinamide could enter the epidermis through the Stratum corneum to the superficial dermis,so as to have the basis for its efficacy,and provide a reference for its efficacy research and formulation application.
