Determination of 18 Polypeptide Compounds in Skin Care Cosmetics by UPLC-MS/MS
A method based on UPLC-MS/MS for the determination of 18 polypeptide compounds in skin care cosmetics was established.The sample was extracted with 50%(in volume fraction)acetonitrile solution.Acetonitrile-0.1%(in volume fraction)formic acid was used as mobile phase for gradient elution at a flow rate of 0.30 mL/min,and the target compounds were separated on a C8 column.A triple quadrupole mass spectrometer operating in positive electrospray ionization mode with multiple reaction monitoring was used.Quantification was carried out by external standard method.Good linear relationships were obtained for 18 polypeptide compounds within their own ranges,and the correlation coefficients(R)were above 0.995.The average recovery rates were in the range of 84.9%-107.4%(n=6),with the relative standard deviations(RSDs)ranging from 1.2%to 7.9%.The limits of detection were 0.01-0.3 mg/kg.The limits of quantitation were 0.03-1.0 mg/kg.Among the 12 batches of skin care cosmetics collected,8 batches detected one or more polypeptide compounds.The method is accurate,sensitive and rapid and it can be used for the qualitative screening and quantitative determination of 18 polypeptide compounds in skin care cosmetics.It also promotes the understanding of actual use and overall risk of protein ingredients in skin care cosmetics and provides technical support for intelligent supervision.
