目的 研究热毒宁注射液与所含绿原酸对RBL-2H3细胞及动物的致敏性.方法 测定热毒宁注射液及绿原酸单次给药刺激RBL-2H3细胞释放的组胺含量;将热毒宁注射液等抗原诱导的特异性IgE大鼠血清预处理RBL-2H3细胞之后,再检测加入热毒宁注射液及绿原酸后细胞释放组胺的含量;利用豚鼠主动全身过敏实验(ASA)及大鼠被动皮肤过敏实验(PCA),观察热毒宁注射液及绿原酸的在体致敏性.结果 热毒宁注射液与绿原酸单次给药不诱导RBL-2H3细胞释放组胺;特异性IgE大鼠血清预处理RBL-2H3细胞后,热毒宁注射液及绿原酸等再次诱导,细胞组胺的释放量无显著性变化;热毒宁注射液以及绿原酸ASA及PCA动物实验结果均为阴性.结论 临床使用剂量下,热毒宁注射液及所含绿原酸不会诱导大鼠及豚鼠发生过敏反应.
Objective To study the sensitization of Reduning injection and the contained chlorogenic acid to RBL-2H3 cells and animals. Methods The content of histamine released from RBL-2H3 cells under the stimulation of single dose of Reduning injection and the contained chlorogenic acid was determined. After RBL-2H3 cells were pretreated by specific IgE rat serum induced by antigen such as Reduning injection and the contained chlorogenic acid, the content of histamine released from the cells added with Reduning injection and the contained chlorogenic acid was determined again; active systemic Anaphylaxis(ASA) experiment on guinea pigs and passive cutaneous anaphylaxis (PCA) experiment on rats were made in order to observe the in-vivo sensitization of Reduning injection and the contained chlorogenic acid. and the contained chlorogenic acid was determined. After RBL-2H3 cells were pretreated by specific IgE rat serum induced by antigen such as Reduning injection and the contained chlorogenic acid, the content of histamine released from the cells added with Reduning injection and the contained chlorogenic acid was determined again; active systemic Anaphylaxis(ASA) experiment on guinea pigs and passive cutaneous anaphylaxis (PCA) experiment on rats were made in order to observe the in-vivo sensitization of Reduning injection and the contained chlorogenic acid.Results Single dose of Reduning injection and chlorogenic acid did not induce RBL-2H3 cells to release histamine; after RBL-2H3 cells were pretreated by specific IgE rat serum, Reduning injection and the contained chlorogenic acid were used for induction again, but the released amount of histamine had no significant changes; ASA and PCA animal experiments results showed that Reduning injection and the contained chlorogenic acid were negative. Conclusion Clinicaldose of Reduning injection and the contained chlorogenic acid will not induce any allergic reaction of rats and guinea pigs.
