Assessments of seed germination characteristics under salt stress for Leymus chinensis germplasms
Leymus chinensis is an elite grass native to temperate grasslands with great potential in restoration of degraded grass-lands and development of artificial pasture.Seed germination and seedling growth of L.chinensis play a key role in the establish-ment of pasture. To assess the application and salt tolerance during seed germination, six L.chinensis germplasms(Caodu No.1,Caodu No.2,Jisheng No.1,Duolun,Hailaer and Changling)were selected to evaluate the seed germination and seedling growth in the absence and presence of 100 mmol L-1 NaCl in the incubation medium. The results showed that Caodu No.1 had the high-est germination percentage, seedling height, root length, total length and simplified vigor index of the whole germplasms treated without NaCl,while Caodu No.2 and Duolun germplasms exhibited the highest germination indexes under salt treatment. The principle component analysis revealed that principle component 1 accounted for more than 90%variation of the used germina-tion percentage(germination number index),total length(seedling growth rate index)and simplified vigor index(growing poten-tial index).Based on the principle component 1,Caodu No.1 and Caodu No.2 had the highest score under conditions with or without salt incubation, respectively, and Duolun germplasm had the highest score in tolerance to salt stress. These results pro-vided useful information for choice of L.chinensis in field sowing, and the methods used in this study could be used to evaluate seed germination and seedling growth of dormant grass species in application to development of pasture.
