Optimization of enzyme production conditions for a high yield amylase producing strain of Weissella
Amylase exists widely in animals,plants and microorganisms,and Weissella has the remarkable ability to produce am-ylase.In this study,an outstanding amylase producing strain W.confusa C13 was selected by starch circle hydrolysis from 3 strains of Weissella,and its biological characteristics were explored.Single factor experiments,Plackett-Burman and steep climb-ing experiments were conducted to improve the condition of enzyme production.Result displayed three most significant factors affecting amylase activity,namely ration of carbon source,pH and the inoculation amount(p<0.05).The optimal enzyme pro-duction conditions were determined as follows,the ratio of skimmed milk powder to sucrose was 4.76∶1,pH 8.07,5.79%inocu-lation amount,3%NaCl,the ratio of soybean meal to yeast was 3∶1,fermentation temperature was 35℃,and fermentation time was 24 hours.Under these conditions,the amylase activity could reach 105.48U/mL,which was 103.43%higher than before optimization,and the optimization effect was significant(P<0.05).The predicted value of the model was basically consistent with the real value(relative error is 3.5%),which provided a theoretical basis for the industrial production of amylase produced by lactic acid bacteria.
