Determination of galuteolin in Liyan gargle by HPLC and the effect of 60 Co-γ radiation on its content
An HPLC method was established to determine the content of galuteolin in Liyan gargle,and the effect of different do-ses of 60 Co-γ radiation was investigated on the content of galuteolin in Liyan gargle.Using Agilent ZORBAX Eclipse XDB-C18(4.6×250 mm,5 μm)as chromatographic column,with acetonitrile-0.1%phosphoric acid(20:80)as the mobile phase,flow rate of 1.0 mL·min-1,and column temperature of 30℃;the detection wavelength was 350 nm,and the injection volume was 10 μL,the content was calculated using the external standard method.Research showed that the specificity of the method for determining the content of galuteolin was good;there was a good linear relationship between the peak area and the range from 1.8954~47.3858 μg·mL-1,with a linear equation of Y =31.13X-15.793 and a correlation coefficient of r =0.9997(n = 6);the average sample recovery rate was 99.73%(n =6);the RSD value for precision was0.58%(n =6),the RSD value for stability was1.26%(n =6),the RSD value for repeatability was 0.87%(n =6),and the RSD for durability was less than3%.After 60 Co-γ radiation,the content of galuteolin in Liyan gargle decreased,and the decrease was positively correlated with the ra-diation dose.This method had strong specificity and high accuracy,and was suitable for the determination of galuteolin content in Liyan gargle.Liyan gargle should not be used for 60 Co-γ radiation sterilization.
