女贞子中多种有效化学成分的同步测定方法研究

建立一种高效液相色谱法同步分析女贞子中咖啡酸、芦丁、槲皮素和熊果酸四种有效化学成分的方法。使用Waters SymmetryshieldTMRP18(4。6 × 250 mm,5 μm)色谱柱,以乙腈-0。1%甲酸水溶液为流动相梯度洗脱,体积流量1。0 mL/min,检测波长范围190-400 nm,柱温30 ℃,进样量10 μL。在此色谱条件下,芦丁、槲皮素、咖啡酸和熊果酸四种成分分离效果良好,在40~250 μg/mL、40~400 μg/mL、80~700 μg/mL和40~400 μg/mL范围内各相关系数均大于0。999,不同浓度条件下芦丁、槲皮素、咖啡酸和熊果酸日内精密度试验的平均RSD值分别为0。41%、0。61%、0。61%、0。86%,日间精密度试验的平均RSD值分别为0。43%、0。52%、0。68%、0。83%,稳定性试验的RSD值分别在0。17%~0。67%、0。54%-0。80%、0。32%~0。87%、0。36%~1。29%,平均回收率分别为 100。81%、97。68%、102。83%、100。01%。该方法精密度高、稳定性良好,可以对女贞子中芦丁、槲皮素、咖啡酸和熊果酸四种有效成分进行同步准确测定,研究结果可为四种有效化学成分的深入研究以及女贞子的开发应用提供研究基础。
Study on the simultaneous analysis of multiple active chemical components in Ligustri Lucidi Fructus
In this study,high performance liquid chromatography(HPLC)method was developed for the simultaneous analysis of four active chemical components,namely caffeic acid,rutin,quercetin and ursolic acid in Ligustri Lucidi Fructus.The separa-tion was performed on a Waters Symmetry shieldTV RP18(4.6~250 mm,5 μm)column with a gradient elution of acetonitrile-0.1%formic acid aqueous solution as the mobile phase,with a volume flow rate of 1.0 mL/min,the detection wavelength in the range of 190~400 nm,the column temperature at 30 degrees Celsius,and the injection volume of 10 μL.Under the chromato-graphic conditions,the four active components,namely rutin,quercetin,caffeic acid and ursolic acid,were well separated and simultaneously analyzed,with the correlation coefficients in the ranges of 40~250 μg/mL,40~400 µg/mL,80~700 μg/mL and 40~400 μg/mL,respectively,which were all greater than 0.999,and the average RSD values of intra-day precision test for rutin,quercetin,caffeic acid and ursolic acid were 0.41%,0.61%,0.61%,0.86%,and the average RSD values of inter-day precision test were 0.43%,0.52%,0.68%,0.83%,and the RSD values of stability test were in the range of 0.17%~0.67%,0.54%~0.80%,0.32%~0.87%and 0.36%~1.29%,and the average recoveries were 100.81%,97.68%,102.83%and 100.01%,respectively.The method had high precision and good stability,and could be used for the simultaneous and accurate determination of the four active components of Ligustri Lucidi Fructus,namely rutin,quercetin,caffeic acid and ur-solic acid,and the results could provide a research basis for the in-depth study of the four active chemical components as well as the development and application of Ligustri Lucidi Fructus.
