Study on the simultaneous analysis of multiple active chemical components in Ligustri Lucidi Fructus
In this study,high performance liquid chromatography(HPLC)method was developed for the simultaneous analysis of four active chemical components,namely caffeic acid,rutin,quercetin and ursolic acid in Ligustri Lucidi Fructus.The separa-tion was performed on a Waters Symmetry shieldTV RP18(4.6~250 mm,5 μm)column with a gradient elution of acetonitrile-0.1%formic acid aqueous solution as the mobile phase,with a volume flow rate of 1.0 mL/min,the detection wavelength in the range of 190~400 nm,the column temperature at 30 degrees Celsius,and the injection volume of 10 μL.Under the chromato-graphic conditions,the four active components,namely rutin,quercetin,caffeic acid and ursolic acid,were well separated and simultaneously analyzed,with the correlation coefficients in the ranges of 40~250 μg/mL,40~400 µg/mL,80~700 μg/mL and 40~400 μg/mL,respectively,which were all greater than 0.999,and the average RSD values of intra-day precision test for rutin,quercetin,caffeic acid and ursolic acid were 0.41%,0.61%,0.61%,0.86%,and the average RSD values of inter-day precision test were 0.43%,0.52%,0.68%,0.83%,and the RSD values of stability test were in the range of 0.17%~0.67%,0.54%~0.80%,0.32%~0.87%and 0.36%~1.29%,and the average recoveries were 100.81%,97.68%,102.83%and 100.01%,respectively.The method had high precision and good stability,and could be used for the simultaneous and accurate determination of the four active components of Ligustri Lucidi Fructus,namely rutin,quercetin,caffeic acid and ur-solic acid,and the results could provide a research basis for the in-depth study of the four active chemical components as well as the development and application of Ligustri Lucidi Fructus.
