莲藕多酚氧化酶的催化特性检测

本试验以冰浴研磨离心法制备莲藕多酚氧化酶酶液,用分光光度法系统的研究了pH值、温度、底物浓度、抑制剂对酶活性的影响.结果表明,以邻苯二酚为底物,莲藕多酚氧化酶的最适pH为7.0,最适温度为20℃;KM为0.18mol/L,Vmax为0.900OD/min;抗坏血酸和硫脲为强烈抑制剂;莲藕多酚氧化酶具有底物专一性.
Detection of Catalytic Characteristics of Polyphenol Oxidase in Lotus Root
In this trial , the PPO in lotus root was extracted by ginding with ice-bath and centrifugal method , and the effects of pH , temperature. concentration of aubatrate. and inhibitors on PPO's activiy were investigated by using the spectrophotometric method. The results showed that the optimal pH and tempearture were 7. 0 and 20 ℃ , Km and Vmax of PPO with catechol aubstrate were 0. 18 mol/L and 0. 900 OD/min, resppectively; sodium sulfite and vitamin C were strong inhibitors;the PPO in lotus root had the aubatrate specificity.
