金福菇菌株遗传多样性的ISSR分析

应用ISSR(inter-simple sequence repeat)分子标记技术对13个不同来源的金福菇(Trichooma lobayense Heim.)菌株进行DNA指纹图谱分析,利用筛选出的7条ISSR引物对金福菇基因组DNA进行ISSR-PCR扩增,共扩增出DNA谱带73条,其中多态性谱带72条,多态位点百分率为98.6%,金福菇各菌株间的遗传相似系数变化范围为0.3425~0.8082.聚类分析结果表明,在遗传相似系数为0.5时,可将13个金福菇菌株划为2大类群.
Genetic Diversity of Tricholoma lobayense Revealed by ISSR Analysis
ISSR(inter-simple sequence repeat) molecular markers were used to analyze DNA fingerprintings of 13 Tricholoma lobayense wildtype suains originating from different locations worldwide.A total of 73 bands were amplified using 7 ISSR primers,of which 72 bands were polymorphic.Perccentage of polymorphic bands was 98.6%.ISSR similarity coefficients ranged from 0.3425 to 0.8082.Cluster analysis further demonstrated that the 13 strains were clustered into two groups at the coefficient of 0.5.
