Genetic Diversity and DNA Fingerprint of Pigmented Potato (Solanumtuberosum L.) Based on ISSR Markers
Genetic polymorphism and ISSR fingerprint of 10 pigmented potato varieties native to Yunnan province were analyzed based on ISSR molecular marker.Fifteen ISSR primers with good polymorphism were selected from seventy ISSR primers.The result showed that 101 bands of ISSR markers were amplified with 15 suitable primers,of which 89 bands were polymorphic,the percentage of polymorphism bands reached 89.24 %,average polymorphic loci of primer was 5.93 and polymorphism information content varied from 0.711 to 0.910.The genetic coefficients (GS) of 10 varieties ranged from 0.503 to 0.919.10 varieties by UPMGA analysis could be clustered into four groups,of which GS between S03-2677 and S03-2685 were the highest,which indicated that there was a very close genetic relationship for two species,but Ziyun1 and S06-277 were the lowest.A core primers AW67776 was screened out to draw a fingerprint of pigmented potato cultivars.It was concluded that 10 pigmented potato varieties were rich in genetic diversity,which could provided a molecular basis for pigmented potato breeding.
