Analysis of Genetic Diversity and Construction of Molecular Fingerprinting with SSR Markers for Ancient Tea Germplasms in Guizhou
In order to effectively conserve and reasonably utilize the ancient tea germplasms in Guizhou,the genetic diversity and molecular fingerprinting of 40 ancient tea plants,collected from the various ecological areas of Guizhou,were analyzed using EST-SSR marker.The resuits showed that total 76 alleles were found using 20 SSR markers with a mean value of 3.8.The average values of PIC and Shannon's information index were 0.462 and 0.936 respectively,indicating the tested germplasms with moderate levd of diversity.The occurrence frequency of 76 alleles differed largely,and it ranged from 3.57 % to 89.29 %.The Nei's genetic distance (D) of tested germplasms varied from 0.075 to 0.875.These germplasms at the D value of 0.16 were clustered into six categories,which included three single categories and three multiple categories.All tested ancient tea plants were identified by four core markers,and the molecular fingerprinting of each ancient tea plant was constructed using the 18-digit number obtained from allele band type.
