食用玫瑰花瓣可培养内生细菌的分离及多样性分析

采用稀释涂布平板法,从食用玫瑰花瓣中分离到219株内生细菌.利用细菌菌落表征性状和16S rDNA序列对这些内生细菌进行了多样性分析.通过数值分析比较了8个菌落形态表征性状,以类平均连锁聚类法的方式进行聚类分析,在3.60的水平上可分成6个表观群.16S rDNA序列分析表明219株内生细菌可分为10个类群.经测序比对分析表明,这219株内生细菌分别与G enBank中3类细菌中的10个已知种相似性达到98.51% ~100%.其中约氏不动杆菌(Acinetobacter johnsonii)、嗜麦寡养食单胞菌(Stenotrophomonas maltophilia)、Enhydrobacter aerosaccus是食用玫瑰花瓣可培养内生细菌的优势种群.
Study on Diversity of Cultivable Endophytic Bacteria Isolated from Petals of Edible Roses
219 endophytic bacteria of Yunnan edible roses were isolated from petals by the method of grinding separation.A preliminary analysis was made on the diversity of the isolates by morphological characteristics and molecular classification methods based on partial 16S rDNA sequences,and the analytical comparison was also made between them.219 bacteria isolates were separated with 8 different morphological characteristics,and were grouped into 6 clusters at the level of 3.60 with numerical classified based on the unweighted pair group method with arithmetic averages algorithm,and they were clustered into 10 molecular groups based on partial 16S rDNA sequences.The result of sequence analysis of the 16S rDNA showed the 98.51%-100 % homology with 10 species in the G enBank.The dominant bacterial groups from petals of edible roses are Acinetobacter johnsonii,Stenotrophomonas maltophilia,Enhydrobacter aerosoccus.
