Effects of drought stress on expression of genes related to starch and sucrose metabolic pathways in taro corms
[Objective]The study aimed to investigate the molecular mechanism of taro corm in response to drought stress,especially the effect of drought stress on the expression of genes related to starch and sucrose metabolic pathways,in order to provide a basis for the selection and breeding of drought-tolerant varieties.[Method]Using Lipu taro 1 as test material,transcriptome sequencing was performed on taro corms un-der drought stress,and the differentially expressed genes obtained were analyzed by GO and KEGG enrichment,to further screen enzyme genes related to starch and sucrose metabolism pathways,and to analyze the expression patterns of these enzyme genes under drought stress.[Result]Taro corms were screened for 1983 differentially expressed genes under drought stress,including 373 up-regulated and 1610 down-regulated genes.GO enrichment analysis showed that differentially expressed genes were most enriched in biological processes for cellular processes and metabolic processes,in cellular components for cellular anatomical entity,and in molecular functions for catalytic activity and binding.KEGG enrichment analysis showed that the top enriched pathways were metabolic pathways,nitrogen metabolism,ascorbate and al-darate metabolism.The differentially expressed genes of starch and sucrose metabolic pathways were counted,and a total of 26 differentially expressed genes involving 11 enzymes were screened,of which 5 were up-regulated expressed genes and 21 were down-regulated expressed genes.Under drought stress,genes for starch synthase,hexokinase,fructokinase,UTP-glucose-1-phosphate uridylyltransferase,α-amylase,en-doglucanase,[3-glucosidase and β-fructofuranosidase were expressed in a down-regulated manner;Genes for UDP-glucose glucophosphohydro-lase and trehalose-6-phosphate synthase/phosphatase were up-regulated;And genes for sucrose synthase and β-furanofuranosidase were ex-pressed in both up-regulated and down-regulated manners.[Conclusion]Expression of genes related to starch and sucrose metabolic pathways was affected by drought stress.Genes involved in the regulation of starch metabolism and alginate biosynthesis were down-regulated,whereas genes involved in the regulation of sucrose metabolism were both up-and down-regulated.
