Correlation analysis between LEP,LEPR gene polymorphism and body condition score in sheep
[Objective]The study aimed to analyze the single nucleotide polymorphisms(SNPs)of LEP and LEPR genes and their correlation with body condition score(BCS)in Texel x Kazak hybrid(Teha)ewes.[Method]The missense mutations in exon 18 of LEP and LEPR were screened by PCR sequencing technology,and the SNPs were genotyped by KASP typing method,and the association between the genoty-ping results and their BCS was analyzed.[Result](i)The results showed that one SNP site,rs1093355763,was found in the LEP gene,and three SNPs sites,rs428867159,rs412929474 and g.2770 G>A,were found in the LEPR gene.All of them belong to exon missense muta-tions,rs428867159 and rs412929474 belonged to moderate polymorphism(0.25<PIC<0.5),and rs1093355763 and g.2770 G>A be-longed to low polymorphism(PIC≤0.25).All of them were in Hard-Weinberg equilibrium(P≤0.05).(ii)In the genotype effect of rs1093355763,the body weight of CA genotype was significantly higher than that of CC genotype at the middle stage of lactation(P<0.05,the samw as below).Among the genotype effects of rs428867159,the BCS of TT genotype was significantly higher than that of CC genotype and CT genotype at lambing stage.In the genotype effect of g.2770G>A locus,the BCS of GA genotype was significantly higher than that of GG genotype at 120 days of pregnancy,and the BCS of GG genotype was significantly higher than that of GA genotype at weaning stage.The body weight of GA genotype was significantly higher than that of GG genotype at 120 days of pregnancy.[Conclusion]The rs1093355763,rs428867159 and g.2770 G>A loci in LEP and LEPR genes can be used as molecular markers for BCS and body weight of Teha ewes in sheep breeding.
