Identification and analysis of bHLH transcription factors family in Docynia delavayi
[Objective]The study aimed to identify and analyze the Docynia delavayi bHLH(DdbHLH)transcription factors(TFs)family and to screen for the DdbHLH TFs related to anthocyanin biosynthesis in the pericarp.[Method]In the study,the family of DdbHLH TFs was identified based on transcriptomic data from the red and green pericarp of D.delavayi,and their bHLH conserved structural domains,protein physicochemical properties,phylogenetic relationships,as well as gene expression were analyzed by bioinformatics tools.[Result]The results showed that a total of 177 DdbHLH TFs family members were identified in D.delavayi with amino acids length ranging from 69 to 699 aa,rel-ative molecular weight ranging from about 7.965 to 75.419 kDa,and an average theoretical isoelectric point of 6.93,most of them belonging to unstable hydrophilic proteins.Phylogenetic relationships showed that 15 subfamilies of DdbHLH TFs co-clustered with Arabidopsis AtbHLH TFs and 17 DdbHLH TFs clustered into anthocyanin-related subfamily 2.Combining the expression of DdbHLH genes in red and green peri-carp,11 members with high expression in red pericarp were preliminarily screened,and differential expression analysis was carried out on the 11 members,and 2 DdbHLH genes that may be involved in the regulation of anthocyanin biosynthesis were finally obtained.RT-qPCR results showed that the relative expression levels of these 2 DdbHLH genes were basically consistent with the transcriptome sequencing,which con-firmed the reliability of the high-throughput sequencing results.[Conclusion]In the study,177 members of the DdbHLH TFs family are iden-tified from the transcriptome data of the red and green pericarps of D.delavayi.The two members(DdbHLH68& DdbHLH 150)are screened out and are found to be associated with anthocyanin biosynthesis in the pericarp,which lays a foundation for the study of DdbHLH TFs family in the regulation of the pericarpcolor of D.delavayi.
