Metabolomic analysis and identification of sterile related target metabolites in sterile flowers of tea plants
[Objective]The present paper aimed to reveal the metabolic characteristics of sterile and fertile flowers,providing targeted metab-olites for studying the formation mechanism of sterile flowers,and thus providing relevant basis for exploring the formation mechanism of ster-ile flowers in tea plants.[Method]The study used the LC-QTOF platform to conduct wide-targeted metabolome detection on completely ster-ile flower tea plant('Yuncha Yirui',ZDH)and its fertile parents('Fuding Dabaicha'and'Foxiang 2',FDDB and FX).Different bioin-formatics software and websites were used to perform principal component analysis,orthogonal partial least squares discriminant analysis,clus-ter analysis,and KEGG pathway analysis on the metabolites,and to analyze the differential metabolites between ZDH,FDDB and FX.[Re-sult]Qualitative and quantitative metabolomic analyses of 9 samples(each experimental material contains 3 biological replicates)identified a total of 963 metabolites.By differential metabolite analysis,a total of 544 differential metabolites(metabolites duplicated between different differential combinations were counted only once)were identified in the three combinations of FX-vs-FDDB,FX-vs-ZDH,and FDDB-vs-ZDH,and the numbers of differential metabolites in different combinations were 373,410 and 224,respectively.This indicated that the metab-olites differed significantly between ZDH and the parents,and the number of metabolites that differed between ZDH and the parents was sig-nificantly higher than the number of metabolites that differed between the parents.Among the 544 differential metabolites,222 components belonged to the differential substances shared by ZDH with FX and FDDB,but there was no difference between FX and FDDB.Nearly one-third(69/222)of the 222 important differential metabolites belonged to flavonoids(kaempferol-associated glycosides,quercetin and many of its glycosides,apigenin-associated glycosides,etc.),saccharides(stachyose,glucose,and fructose,etc.),and hormone-associated substances(methoxyindoleacetic acid,5'-glucosyloxyjasmanic acid,and salicylic acid-2-O-glucoside,etc.),and the differentials in these metabolites may be involved in the formation of ZDH sterility.[Conclusion]There are significant differences in the metabolic components between sterile and fertile flowers,and flavonoids,sugars,and hormone related substances,especially kaempferol derivatives,auxin,and jasmonic acid may be involved in the formation of tea plant flower infertility phenotype.
