Therapeutic Effect of Qingzhuan Dark Tea Aqueous Extract on Constipated Irritable Bowel Syndrome in Rats
Objective To explore the effect of Qingzhuan Dark Tea aqueous extract on constipated irritable bowel syndrome in rats.Methods The 9-year-old Qing Zhuan Dark Tea was crushed,extracted with hot water,and the filtered fluid was concentrat-ed and lyophilized to obtain the Qingzhuan Dark Tea aqueous extract.Female SD rats were randomly divided into control group,model group and Qingzhuan Dark Tea extract group(QDT group,50 mg/kg),with 6 rats in each group.The model of constipation-typeirritable bowel syndrome was established by ice water gavage in 14 days.Then the QDT group was given corresponding drug dose once a day for 14 days,and the control group and the model group were given the equal of normal saline.The body weight,fe-cal pellet number and fecal water content of rats were observed and recorded,the intestinal transit function of rats was detected by charcoal propulsion rate,the pathological changes of colon tissue were observed by HE staining,and the expression of serotonin pro-tein in serum and5-HT3 R protein in colon tissues was detected.Results Compared to the control group,the rats in the model group exhibited a reduction of the body weight gain trend,a lower number of fecal pellets,and decreased fecal moisture content,dimin-ished intestinal motility.Histological analysis using Hematoxylin-Eosin(HE)staining revealed no organic lesions in the colon tis-sue,while the levels of serotonin 5-HT were significantly elevated and the expression of 5-HT3 R was notably reduced.In contrast,when compared to the model group,the rats in the QDT group demonstrated a tendency for increased body weight gain and a higher number of fecal pellets,along with significantly enhanced fecal moisture content and intestinal motility.Additionally,the extent of inflammatory infiltration in the colon was reduced and the levels of 5-HT were decreased.Conclusion The Qingzhuan Dark Tea a-queous extract can alleviate the constipation symptoms of constipation-type irritable bowel syndrome in rats,and the mechanism may act on the 5-HT signaling pathway,which reduce the level of 5-HT,and induce the up-regulation of 5-HT3 R expression in the colon.
