猪繁殖与呼吸综合征(Porcine reproductive and respiratory syndrome,PRRS)是影响全球养猪业的主要疾病之一.猪繁殖与呼吸综合征病毒(PRRSV)的突变率是RNA病毒中最高的.为了解其分子特征和遗传变异,对陕西和河南地区猪场采集的252 份疑似样本采用RT-PCR检测技术和生物信息学软件进行了ORF5 基因和Nsp2 基因序列比对和遗传变异分析.结果显示:252 份样本中阳性样品为39 份,阳性率为 15.6%.对 39 份阳性样品克隆测序获得 10 条Nsp2基因阳性样品序列和5 条ORF5 基因阳性品序列;5 株PRRSV ORF5 基因毒株与GenBank中 26 株PRRSV参考毒株的ORF5 基因之间的核苷酸序列同源性为79.7%~99.7%,10 株PRRSV毒株Nsp2 基因的核苷酸序列同源性为 38.2%~92.2%,其中5 株ORF5 基因的GP5-5、GP5-7、GP5-10 毒株为PRRSV-2 型谱系 1 的毒株,99L、100L毒株为PRRSV-2 型谱系5 的毒株,分别与代表株NADC30 和VR-2332 亲缘关系最近;10 株PRRSV毒株Nsp2 基因均为PRRSV-2 型谱系 1 的毒株,与代表株NADC30 亲缘关系最接近.GP5 蛋白与NSP蛋白均发生不同程度的变异,说明 PRRSV 毒株变异频繁,因此,实时监测 PRRSV株系的遗传变异动态,探索其遗传变异模式,可为疫病防控提供科学参考.
Epidemiological investigation and sequence analysis of reproductive disorder and respiratory syndrome in pig farms in Shaanxi and Henan provinces
Porcine reproductive and respiratory syndrome(PRRS)is one of the major diseases affecting the global swine indus-try.PRRSV has the highest mutation rate among RNA viruses.In order to understand its molecular characteristics and genetic var-iation,252 suspected samples collected from pig farms in Shaanxi and Henan regions were analyzed for ORF5 gene and Nsp2 gene sequence comparison and genetic variation analysis using RT-PCR assay and bioinformatics software in this study.The re-sults showed that 39 out of 252 samples were positive,with a positive rate of 15.6%.Sequencing of the 39 positive samples yiel-ded ten Nsp2 gene-positive samples and five ORF5 gene-positive samples.The nucleotide sequence homology between the five PRRSV ORF5 gene strains and the ORF5 gene of the 26 PRRSV reference strains in GenBank ranged from 79.7%to 99.7%,and that between the ten PRRSV strains and the ORF5 gene ranged from 38.7%to 99.7%,and that between the ten PRRSV strains and the ORF5 gene ranged from 38.7%to 99.7%.Nucleotide sequence homology ranged from 38.2%to 92.2%;the ho-mology of the nucleotide sequences of the ORF5 genes of the five strains ranged from 85.1%to 99.8%,among which the five strains with the ORF5 genes of strains GP5-5,GP5-7,and GP5-10 were strains of the PRRSV-2 lineage 1,and strains 99L and 100L were strains of the PRRSV-2 lineage 5,which were identical to representative the Nsp2 gene of the 10 PRRSV strains is a strain of PRRSV-2 lineage 1,99L and 100L strains are strains of PRRSV-2 lineage 5,which are closest to the representative strains NADC30 and VR-2332,respectively;the Nsp2 gene of the 10 PRRSV strains is a strain of PRRSV-2 lineage 1,which is the closest to the representative strain NADC30;the GP5 and NSP proteins are mutated to varying degrees,which indicates that the PRRSV strains are frequently mutated,so we need to monitor the dynamics of the PRRSV strains and explore their patterns of genetic mutation in real-time.Therefore,real-time monitoring of the genetic variation of PRRSV strains and exploring their genetic variation patterns can provide scientific references for the prevention and control of epidemics.
