我国猪萨佩罗病毒分子流行病学及检测方法研究进展

猪萨佩罗病毒(porcine sapelovirus,PSV)是一种可引发仔猪腹泻、生长发育迟缓、肌肉震颤等多系统症状的重要病原体,严重影响养猪业的发展.已有研究表明,PSV已在我国多个省市呈现流行趋势,且不同地区的病毒毒株存在一定的差异.由于尚无抗病毒药物和有效疫苗,早期识别和精准诊断对科学防控PSV感染具有决定性的作用.目前,PSV分子检测方法主要分为2 大类:免疫学检测技术和核酸检测技术.免疫学检测技术包括酶联免疫吸附试验和间接免疫荧光技术;核酸检测技术包括逆转录聚合酶链反应(RT-PCR)、多重RT-PCR、巢式RT-PCR、荧光定量PCR、多重荧光定量PCR、环介导等温扩增、重组酶聚合酶扩增以及原位杂交技术等.在此基础上,作者对PSV分子检测方法的优缺点进行了讨论,并对其未来发展方向进行了展望,以期为我国科学防控PSV提供参考依据.
Research progress on molecular epidemiology of porcine sapelovirus in China as well as its detection methods
Porcine sapelovirus(PSV)is an important pathogen that can cause multi-system symptoms such as diarrhea,growth retardation,and muscle tremor in piglets,which seriously affects the development of the pig industry.Previous studies have shown that PSV has shown an epidemic trend in many provinces and cities in China,and there are some differences in virus strains in different regions.Since there are no antiviral drugs and effective vaccines,early identification and accurate diagnosis play a decis-ive role in the scientific prevention and control of PSV infection.Currently,molecular detection methods for PSV can be divided into two main categories:immunological technologies and nucleic acid detection technologies.Immunological technologies include enzyme-linked immunosorbent assay and indirect immunofluorescence assay.Nucleic acid detection technologies include reverse transcription polymerase chain reaction(RT-PCR),multiplex RT-PCR,nested RT-PCR,fluorescence quantitative PCR,multiplex quantitative PCR,loop-mediated isothermal amplification,recombinase polymerase amplification,and in situ hybridization.On this basis,the authors discussed the advantages and disadvantages of the above methods and look forward to their future development directions in order to provide a reference for the scientific prevention and control of PSV in China.
