摘要
以木霉(Trichoderma sp.)为出发菌株,通过紫外诱变与光复活交替处理获得了 1 株高产β-葡聚糖酶突变株UN123.该菌株在摇瓶中的平均酶活大于10 000 U/mL,最适反应温度是40℃,90℃条件下保温处理2h,仍能保持80%以上酶活,最适反应pH是3.0,在pH2.0~7.0 的条件下处理2h后,相对酶活力保持在80%以上且对胃蛋白酶、胰蛋白酶及凝乳蛋白酶具有良好的耐受性.
Abstract
Using Trichoderma sp.As original strain,a mutant strain UN123 with high yield β-glucanase was obtained by UN and photoreactivation.The β-glucanase activity of Trichoderma sp.UN123 in shake flask was above 10 000 U/Ml,its optimum tem-perature was 40℃.The β-glucanase activity of Trichoderma sp.UN123 maintained above 80%even given heat preservation of 2 h under the temperature of 90℃.Its optimum pH was 3.0.The β-glucanase activity of Trichoderma sp.UN123 maintained above 80%under the condition of pH 2.0~7.0 of 2 h and showed god tolerance to pepsin,trypsin,and chymotrypsin.
