Comparative Evaluation of DNATyper? 21 and Verifiler? Plus Kit in Forensic DNA Casework
The DNATyper™ 21 amplification kit is a 5-dye multiplex assay,which is widely used in construction of the Chinese National autosomal DNA database.However,the DNATyper™ 21 kit has not been extensively used in forensic DNA casework.Therefore,this study aimed to elucidate the reliability of the DNATyper™ 21 kit compared with Verifiler™ Plus kit and to assess its applicability on forensic analysis.Standard DNA samples with concentrations of 0.031 25,0.062 5,0.125,0.25,0.5 and 1.0 ng/μL were used for sensitivity testing.DNATyper™ 21 kit and Verifiler™ Plus kit were utilized to detect the DNA extracted from 1056 casework evidence samples and the allele consistency of the same sample was analyzed.The results showed that both kits could successfully detect standard DNA with a template concentration above 0.062 5 ng/μ.L.For casework evidence samples,DNATyper™ 21 kit successfully detected 881 cases out of 1 056,and the detection rate is 83.4%,while Verifiler™ Plus kit successfully detected 892 cases out of 1 056,and the detection rate is 84.5%.The statistical results showed that there was no significant difference in the detection rate between the two kits,and the alleles obtained at the same locus of the same sample were completely consistent.In conclusion,the DNATyper™ 21 is considered to have the capability to be used for STR analysis of casework evidence samples.
