摘要

基于网络药理学研究方法,利用SymMap数据库获得川楝子治疗肝癌的化学成分及靶点基因,从GeneCards数据库获取疾病靶点基因,将川楝子和肝癌的交集靶点导入GeneMANIA数据库获得靶点互作关系,并进行GO和KEGG通路富集分析,结合关键靶点基因在肝癌组织中的表达与预后的相关性分析,筛选到槲皮素等6种主要活性成分,TP53、AKT1、IGF2、CDKN2A等主要靶点,癌症通路、癌症中蛋白聚糖、PI3K-Akt信号通路、乙肝和大肠癌通路等主要调控路径,揭示了川楝子治疗肝癌的机制具有多成分、多靶点和多通路的性质,从整体上为川楝子治疗肝癌提供参考.

Abstract

Based on the network pharmacology research method,the chemical composition and target genes of Melia toosendan fruits for liver cancer were obtained from SymMap database,the disease target genes were obtained from GeneCards database,the intersecting targets of Melia toosendan fruits and liver cancer were imported into the Gene-MANIA database to obtain the target interactions,and GO、KEGG pathway enrichment analyses were carried out.Combined with the correlation analysis between the expression of key target genes in liver cancer tissues and progno-sis,six major active ingredients such as quercetin,major targets such as TP53,AKT1,IGF2 and CDKN2 A,and major regulatory pathways such as the cancer pathway,proteoglycan,PI3 K-Akt signaling pathway,and hepatitis B and colorectal cancer pathway were screened.The effect of Melia toosendan fruits acting on liver cancer are multi-com-ponent,multi-target and multi-pathway.Through the analysis of biological big data and network pharmacology,it provides a reference for the research of Melia toosendan fruits on liver cancer as a whole.
