Effects of platycodin D on programmed death ligand 1 expression and angiogenesis in human colon cancer cells
OBJECTIVE To explore the effect of platycodin D(PLD)on programmed death ligand 1(PD-L1)expression,colony formation,wound healing and angiogenesis of endothelial cells in human colon cancer cells.METHODS MTT assay was used to detect the effect of PLD on the proliferation of human colon cancer cells HCT116.Western blot assay was used to detect the effect of PLD on the expression of PD-L1 protein in HCT116 cells.Cell colony formation experiment was used to observe the effect of PLD on the proliferation ability of HCT116 cells.Scratch assay was used to observe the effect of PLD on cell migration rate.Tubule formation assay was used to observe the effect of PLD on the angiogenesis of endothelial cells.RESULTS PLD could inhibit the proliferation of tumor cells(P<0.001),and inhibit the expression of PD-L1 protein in HCT116 cells in a concentration and time dependent manner.PLD could inhibit the cell colony formation and migration of HCT116 cells and angiogenesis of endothelial cells.CONCLUSION PLD can inhibit the expression of PD-L1 in human colon cancer HCT116 cells,and inhibit the proliferation,migration of HCT116 cells and angiogenesis.
