Protective effect of Gegen Jieji Decoction on renal injury in diabetic mice
OBJECTIVE To investigate the protective effect of Gegen Jieji Decoction on renal injury in diabetic mice.METHODS The 6-week-old SPF male mice were given peritoneal injection of streptozotocin(150 mg/kg)to establish diabetic nephropathy model(fasting blood glucose≥11.1 mmol/L after 72 hours of streptozotocin injection was considered to be successful in making the model),and the normal group(n=7)was given an equal volume of lemonade buffer by peritoneal injection.After the modeling was successful,they were randomly divided into model group,Gegen Jieji Decoction low dose group,high dose group(150,600 mg/kg)and metformin group(0.1 g/kg),7 mice in each group,and were given the corresponding dose of drug by intragastric administration for 6 weeks.Fasting blood glucose,body mass and water intake were observed during administration.The levels of serum superoxide dismutase(SOD),malondialdehyde(MDA),blood urea nitrogen(BUN)and serum creatinine(Scr)were detected after 6 weeks of intragastric treatment.HE staining and PAS staining were used to observe the pathological morphological changes of renal tissue in mice of each group.RESULTS Compared with model group,fasting blood glucose and water intake were significantly decreased(P<0.05),and body mass was significantly increased(P<0.05)in Gegen Jieji Decoction high dose group;serum SOD content was significantly increased(P<0.05),MDA,BUN and Scr levels were significantly decreased(P<0.05);the results of HE staining and PAS staining showed that the degree of glomerular injury,cell arrangement and proliferation were significantly improved.CONCLUSION Gegen Jieji Decoction has protective effect on renal injury of diabetic mice.
