摘要

[目的]探讨Prx V在芝麻素增强吉西他滨诱导的人肺腺癌A549细胞凋亡中的作用机制,旨在为肺癌的治疗提供靶点.[方法]取人肺腺癌A549细胞株体外培养48 h,采用MTT法检测不同浓度吉西他滨及芝麻素作用下的A549细胞的存活率,并计算半数抑制浓度.[结果]吉西他滨、芝麻素作用于人肺腺癌A549细胞的半数抑制浓度分别为7、45 μmol/L.划痕实验结果显示联合用药组划痕修复率显著低于单用药组(P<0.05,P<0.01).TUNEL染色结果显示联合用药组凋亡细胞数显著高于吉西他滨组和芝麻素组(P<0.01).JC-1荧光探针标记检测结果显示联合用药组细胞中线粒体膜电位降低幅度显著高于吉西他滨组和芝麻素组(P<0.05).荧光探针DCFH-DA检测结果显示吉西他滨组和芝麻素组细胞ROS水平显著低于联合用药组.蛋白印迹实验结果显示吉西他滨和芝麻素均可抑制A549细胞中Prx V表达,联合用药组抑制作用更为显著.[结论]吉西他滨和芝麻素均可抑制人肺腺癌A549细胞的增殖和迁移,且芝麻素可增强吉西他滨的上述作用;吉西他滨可降低Prx V表达,从而减弱Prx V对人肺腺癌A549细胞的保护作用,芝麻素可增强吉西他滨的这一作用;吉西他滨和芝麻素可降低人肺腺癌A549细胞内Prx V表达,升高细胞内ROS水平,降低线粒体膜电位,从而使细胞发生凋亡.

Abstract

OBJECTIVE To investigate the mechanism of Prx V in sesamin enhancing gemcitabine-induced apoptosis of human lung cancer A549 cells,so as to provide a therapeutic target for the treatment of lung cancer.METHODS Human lung cancer cell line A549 was selected and cultured in vitro for 48 hours.The survival rate of A549 cells under the action of different concentrations of gemcitabine and sesamin was measured by MTT method,and the half inhibitory concentration was calculated.RESULTS The IC50 of gemcitabine and sesamin on human lung cancer A549 cells were 7 and 45 μmol/L,respectively.The scratch experiment results showed that the scratch repair rate of the combination drug group was significantly lower than that of the single drug group(P<0.05,P<0.0l).TUNEL staining results showed that the number of apoptotic cells of the combination drug group was significantly higher than that of gemcitabine and sesamin group(P<0.01).The results of JC-1 fluorescent probe labeling showed that the reduction of mitochondrial membrane potential in the combination drug group was significantly higher than that in the gemcitabine group and sesamin group(P<0.05).Fluorescent probe DCFH-DA test showed that the ROS levels in cells of the gemcitabine group and sesamin group were significantly lower than that in the combination drug group.The results of the Western blot experiment showed that both gemcitabine and sesamin could inhibit the expression of Prx V in A549 cells,and the inhibition was more significant in the combination drug group.CONCLUSIONS Both gemcitabine and sesamin can inhibit the proliferation and migration of human lung cancer A549 cells,and sesamin can enhance the above effects of gemcitabine.Gemcitabine can reduce the expression of Prx V,thereby weakening the protective effect of Prx V on lung cancer cell A549.Sesame can enhance this effect of gemcitabine.Gemcitabine and sesamin can reduce the expression of Prx V in human lung cancer A549 cells,increase intracellular ROS level,and decrease mitochondrial membrane potential,thus leading to cell apoptosis.
