摘要

目的:优化春榆中总黄酮的提取技术后,利用紫外-可见光谱法(UV-Vis)检测春榆中总黄酮的含量.方法:春榆总黄酮提取率的效果以温度,乙醇体积分数和时间通过正交试验法,确定最优的春榆提取技术,通过UV-Vis检测春榆中总黄酮含量.结果:确定以50%的乙醇、温度75℃、时间100 min为春榆总黄酮最优提取条件,故总黄酮提取率为9.69%.结论:乙醇回流提取法成本低廉、易操作,正交试验设计法优化春榆总黄酮提取工艺条件,简单易行、重复性佳,为春榆总黄酮的研究提供了一定参考依据.

Abstract

OBJECTIVE To determine the content of total flavonoids in Ulmus davidiana var.japonica by ultraviolet-visible spectroscopy(UV-Vis)after optimizing the extraction technology of total flavonoids from Ulmus davidiana var.japonica.METHODS The optimal extraction technology of Ulmus davidiana var.japonica for total flavonoids extraction rate was determined by orthogonal test based on temperature,ethanol volume fraction and time.The total flavonoid content in Ulmus davidiana var.j aponica was detected by UV-Vis.RESULTS The optimal extraction conditions for total flavonoids in Ulmus davidiana var.j aponica were 50%ethanol,a temperature of 75℃,and a duration of 100 minutes.Under these conditions,the extraction rate of total flavonoids was 9.69%.CONCLUSION Ethanol reflux extraction method is cheap and easy to implement.The orthogonal experimental design method optimizes the extraction conditions for total flavonoids in Ulmus davidiana var.j aponica,which is simple,feasible,and highly reproducible,and provides a valuable reference for research on total flavonoids in Ulmus davidiana var.j aponica.
