从陇南花椒根围土壤中筛出3种对腐皮镰刀菌具有较强拮抗效果的生防菌株,通过形态学结合分子生物学方法分别鉴定为枯草芽孢杆菌W-5、贝莱斯芽孢杆菌T-1、贝莱斯芽杆菌W-1.试验确定3种拮抗菌的发酵条件参数后,以3种拮抗菌的发酵液的混合液进行复合微生物生防菌剂的研发,得到复合菌剂的最佳载体为硅藻土,吸附量为2.5 mL/g;最佳分散剂为木质磺酸钠,添加量为6%;最佳紫外保护剂为海藻酸钠,添加量为8%.经测定复合菌剂具有广泛的抑菌活性,对腐皮镰孢菌、梅毒镰刀菌、松针刺盘孢菌、禾谷镰刀菌、核桃炭疽病病原菌菌丝的抑制率分别为83.3%、79.3%、74.7%、89%、83.7%,复合菌剂稀释2000倍时对花椒根腐病病原菌菌丝的抑制率在95%以上;其pH值为6.75,含水量0.25%,有效活菌数3.17×108CFU/g,保质期 0.8~1 年,达到 GB 20287-2006标准.
Preparation of a Compound Microbial Agent GF-1 for Prevention and Treatment of Zanthoxylum Root Rot Disease
Three biocontrol strains with strong antagonistic effect against Fusarium fusarium were screened from the soil around the root of Zanthoxylum in Longnan,and identified as Bacillus subtilis W-5,Bacillus Bellescens T-1 and Bacillus Bellescens W-1 by morphological and molecular biological methods.After the fermentation conditions of 3 kinds of antagonistic bacteria were deter-mined,the composite microbial biocontrol agent was developed with the mixture of 3 kinds of antagonistic fermentation liquid.The best carrier of the composite bactericide is diatomite,and the adsorption capacity is 2.5 mL/g.The best dispersant is sodium ligno-sulfonate,the addition amount is 6%;The best UV protective agent is sodium alginate,the addition amount is 8%.The results show that the antibacterial activity of the compound bacteriological agent is 83.3%,79.3%,74.7%,89%and 83.7%,respectively,against Fusarium dermatitis,Fusarium syphilitica,Fusarium piniculata,Fusarium graminea and anthracnose of walnut.When diluted 2000 times,the inhibition rate of mycelium of pathogenic bacteria of Zanthoxylum root rot is over 95%.Its pH value is 6.75,water con-tent is 0.25%,effective viable bacteria number 3.17×108 CFU/g,shelflife of 0.8~1 years,up to GB 20287-2006 standard.
