葛根异黄酮对去卵巢大鼠抗骨质疏松的疗效

目的::观察葛根异黄酮治疗去卵巢大鼠骨质疏松的效果。方法:选用健康3月龄SD雌性大鼠50只,随机分为5组:假手术组、模型对照组、低、中、高剂量葛根异黄酮组;假手术组切除卵巢旁脂肪组织,其余各组均切除双侧卵巢,建立去卵巢大鼠动物模型。按25、50、100 mg/kg 3个剂量葛根异黄酮分别给去卵巢大鼠灌胃6个月,第3、6个月测量体重,并于实验结束后处死大鼠,检测血清钙、磷、碱性磷酸酶水平和股骨中心、远心骨密度。结果:中、高剂量治疗组动物较模型对照组在第3、6个月体重均降低,在第6个月血清钙明显升高,血清磷和碱性磷酸酶水平明显降低。高剂量治疗组动物较模型对照组在第6个月股骨中心和远心骨密度均明显升高。结论:葛根异黄酮长期治疗可升高血清钙水平,降低血清磷和碱性磷酸酶水平,改善骨密度,具有抗骨质疏松的作用。
Object:To observe the effect of Totalisoflavone on osteoporosistreatment inovariectomized rats.Methods:We randomly distributed 50 healthy and fe-male rats with 3-month of age into 5 groups:sham operation group, model control group and Total isoflavonespueraria groups with low, medium and high-dose ad-ministration. Thesham group was removed adipose tissuesaroundovarian, the others were all ovariectomizedrats.The ovariectomizedrats were model controls, or were intragastric administrated with Totalisoflavone at the dose of 25, 50 or 100 mg/kg. In the third and sixth month, weights of rats were observed. In the six month, the levels of Ca+, P and alkaline phosphatase (ALP) in serum, and the bone mineral density (BMD) in both center and far-end of femur were tested.Results:The weights of medium and high-dose groups were lower than the model controls in the third and sixth month. Compared with the model controls, the levels of Ca+in serum were increased, and the levels of P and ALP in serum were decreased in medium and high-dose groups in the sixth months. The BMDs in both center and far-end of femur were higher in high-dose group than the model controls in the sixth months. Conclusions: Treatment with Totalisoflavone in a long term could increase the level of Ca+in serum, decrease the level of P and ALP in serum, improve BMD, showing an effect of anti-osteoporosis.
