河南植烟区烟草黑胫病菌生理小种的鉴定及遗传多样性分析

为了探索河南主要烟草种植区烟草黑胫病菌生理小种及遗传分化关系,采用实验室组织培养瓶鉴定法对从河南烟叶产区分离到的28株黑胫病菌进行了生理小种鉴定,同时利用RAPD熔解曲线分析法(McRAPD)对28株烟草黑胫病菌进行了遗传多样性分析。结果表明:①从28个烟草黑胫病菌分离株中准确鉴定出25株1号生理小种,占供试菌株的89.2%,其余3株黑胫病菌的生理小种类型尚未确定;②采用非加权组平均法(UPGMA),以遗传距离0.79将供试的28个菌株划分为6个遗传聚类组,除部分同一或临近烟区的菌株被划分为同一遗传聚类组外,大多数菌株的遗传分化与地理位置远近没有直接的相关性,与生理小种类型的划分也没有直接的相关性。
Genetic diversity analysis and physiological race identification of Phytophthora parasitica var. nicotinanae in Henan tobacco planting areas
To identify the physiological races and genetic diversity of Phytophthora parasitica var. nicotinanae in major tobacco planting areas of Henan Province, twenty-eight physiological races were identified by a laboratory tissue culture method and the genetic diversity analysis of twenty-eight pathogens was conducted by melting curve of RAPD (McRAPD). The results showed that: 1) Twenty-five P. parasitica var. nicotinanae isolates were accurately identified as the 1st physiological race, accounting for 89.2% of tested strains, and the other three strains could not be determined at the present. 2) Tested strains were clustered into six groups according to unweighted pair group method with arithmetic average (UPGMA) and the genetic similarity coefficient of 0.79. Except that several strains clustered into the same group were from the same area or close areas, the genetic variation of most strains did not directly correlated to geographical origins and physiological race types.
