Genetic diversity analysis and physiological race identification of Phytophthora parasitica var. nicotinanae in Henan tobacco planting areas
To identify the physiological races and genetic diversity of Phytophthora parasitica var. nicotinanae in major tobacco planting areas of Henan Province, twenty-eight physiological races were identified by a laboratory tissue culture method and the genetic diversity analysis of twenty-eight pathogens was conducted by melting curve of RAPD (McRAPD). The results showed that: 1) Twenty-five P. parasitica var. nicotinanae isolates were accurately identified as the 1st physiological race, accounting for 89.2% of tested strains, and the other three strains could not be determined at the present. 2) Tested strains were clustered into six groups according to unweighted pair group method with arithmetic average (UPGMA) and the genetic similarity coefficient of 0.79. Except that several strains clustered into the same group were from the same area or close areas, the genetic variation of most strains did not directly correlated to geographical origins and physiological race types.
