HPLC Content Determination and Extraction Process Optimization of Scopolamine in Herba Hedyotids Chrysotridhae
Objective:To establish an HPLC method for the determination of scopolamine in Her ba Hedyotids Chrysotridhae and optimize its extraction process.Method:The content of scopoletin was determined using Waters Symmetry-C18(4.6 mm)× 250 mm,5 μm)Chromatographic column,acetonitrile 0.1%phosphoric acid wa-ter as mobile phase,detection wavelength of 345 nm,flow rate of 0.8 mL/min,injection volume of 10μL.Using the ultrasonic extraction method,based on a single factor experiment using ethanol concentration(A),ultrasonic time(B),ultrasonic power(C),and solid-liquid ratio(D)selected as the evaluation factors.The content of scopolamine was used as the evaluation index,and the Box Behnken response surface method was used to optimize the extraction process of scopolamine in Herba Hedyotids Chrysotridhae.Result:The optimal extrac-tion process is ethanol concentration of 74%,extraction time of 45 minutes,ultrasonic power of 250 W,and liq-uid-solid ratio of 25∶1.Under these conditions,the content of scopolamine is 81.97 μg/g,which showed no sig-nificant difference from the predicted value of 81.41 μg/g.Conclusion:The HPLC method established in this ex-periment can be used for the determination of scopolamine in Herba Hedyotids Chrysotridhae.The optimized ex-traction process is stable,reliable,simple,and feasible.
