江枳壳果胶的提取方法及其工艺优化

为考察水提醇沉法和酸提醇沉法提取江枳壳果胶的提取率,并探究酸提醇沉法提取江枳壳果胶的工艺条件,分别采用两种提取方法提取江枳壳果胶,计算提取率,针对酸提醇沉法进行单因素试验,以单因素试验结果为基础,通过Box-Behnken响应面试验设计方案,确定江枳壳果胶多糖的最佳提取工艺.结果表明,采用水提醇沉法提取江枳壳果胶多糖的提取率为0.92%,采用酸提醇沉法提取江枳壳果胶多糖的提取率为17.18%.采用酸提醇沉法提取江枳壳果胶多糖的最佳提取工艺如下:液料比22∶1(mL/g),pH为0.75,浸提时间为100 min,浸提温度81 ℃,理论提取率为23.34%.依据此提取条件进行3次平行试验,得到江枳壳果胶多糖提取率为23.28±0.51%,与预测值相符.研究结果表明,酸提醇沉法比水提醇沉法提取江枳壳果胶多糖的提取率高.响应面法优化江枳壳果胶提取工艺,方法可行,操作简单,提高了果胶提取率.
Extraction Method and Process Optimization of Jiangxi Fructus Aurantii Pectin
In order to investigate the extraction rate of pectin from Jiangxi Fructus Aurantii by water extraction and alcohol precipitation method and acid extraction and alcohol precipitation method,and to explore the technological conditions of extracting pectin from Jiangxi Fructus Aurantii by acid extraction and alcohol precipitation method,two extraction methods were used to extract pectin from Jiangxi Fructus Aurantii,and the extraction rate was cal-culated.The single factor experiment was carried out for the acid extraction and alcohol precipitation method.Based on the results of single factor experiment,and the optimum extraction process of pectin polysaccharide from Jian-gxi Fructus Aurantii was determined by Box-Behnken response surface experimental design scheme.The results showed that the extraction rate of polysaccharide extracted by water extraction and alcohol precipitation was 0.92%,and the extraction rate of polysaccharide extracted by acid extraction and alcohol precipitation was 17.18%.The optimum extraction process of pectin polysaccharide from Jiangxi Fructus Aurantii by acid extraction and alcohol precipitation was as follows:liquid-solid ratio 22:1(mL/g),pH0.75,extraction time 100 min,extraction temperature 81 ℃,the theoretical extraction rate was 23.34%.According to this extraction condition,three parallel experiments were carried out,and the extraction rate of pectin polysaccharide from Jiangxi Fructus Aurantii was 23.28±0.51%,which was consistent with the predicted value.The results showed that the extrac-tion rate of acid extraction and alcohol precipitation method was higher than that of water extraction and alcohol precipitation method.The optimization of extraction process of Jiangxi Fructus Aurantii through response surface methodology is feasible,with simple operation and improved pectin extraction rate.
