Effects of sanguis draconis flavones on the HMGB1/PI3K pathway in rat cardiomyocytes after ischemia/reperfusion injury
Objective To observe the interventional effects of sanguis draconis flavones(SDF)on a rat myocardial ischemia/reperfusion(I/R)model and investigate its effect on HMGB1/PI3K pathway.Methods Thirty-two healthy male SD rats were randomly divided into Control,Sham,I/R and SDF groups,with 8 rats in each group.To establish an I/R model,we performed ligation of the mid-left anterior descending coro-nary artery.Creatine kinase isoenzyme(CK-MB)levels were measured by enzyme-linked immunosorbent assay(ELISA),and the pathological changes were observed via hematoxylin and eosin staining.Triphenyltetrazoli-um chloride staining was used to accurately assess myocardial infarction size.The expression levels of HMGB1 and P13K in rat myocardium were detected by Western Blot technique.Results ①Compared with the Control and Sham groups,CK-MB expression,myocardial infarct area,and HMGB1 and PI3K protein levels were sig-nificantly increased in the I/R and SDF groups(P<0.05).②Histological examination revealed intact myo-cardial architecture and orderly arranged cardiac muscle fibers in the Control and Sham groups.The cardiomyo-cytes in group I/R were disordered,with interstitial edema and some cells showed necrosis.In addition,the in-filtration of the inflammatory cells can also be observed.Compared with the I/R group,the myocardial struc-ture destruction in SDF group was significantly alleviated,with only mild cell death,neater fiber arrangement,and a small amount of neutrophil infiltration observed.Conclusion Pretreatment with sanguis draconis fla-vones(SDF)can modulate the HMGB1/PI3K signaling pathway,thereby exerting a protective effect against myocardial ischemia/reperfusion injury.
