Protective effects of Panax notoginseng saponins on OGD/R-induced endothelial cell barrier and function through PPARα/Nrf2 activation
Objective This study was implemented to investigate the protective role of Panax notoginseng saponins(PNS)against OGD/R-induced damage to endothelial cell barrier and function and to elucidate the un-derlying mechanism involving PPARα/Nrf2 signaling.Methods To establish an ischemia-reperfusion injury model,bEnd.3 cells were induced by OGD/R.Cells were divided into Control,OGD/R,high-dose PNS(OGD/R+PNS 400μg/mL),PPARα inhibitor(OGD/R+PNS 400μg/mL+PPARα inhibitor)and Nrf2 inhibi-tor(OGD/R+PNS 400μg/mL+NRF2 inhibitor)groups.The cell viability was detected using CCK-8 and LDH assay was used for the measurement of lactate dehydrogenase(LDH)level.The protein expressions of ZO-1,claudin-5,occludin were evaluated with Western Blot.Wound healing and transwell assays were em-ployed to detect migration and invasion of bEnd.3 cells.Besides,the tube forming ability of bEnd.3 cells was assessed using tube formation assay.Results PNS alleviated OGD/R-induced bEnd.3 cell viability impair-ment,attenuated endothelial cell barrier damage,inhibited cell migration and invasion and ameliorated angio-genic damage through activation of peroxisome proliferator-activated receptor alpha/nuclear Factor Erytroid 2-related factor 2(PPARα/Nrf2)signaling.Conclusion PNS exerts protective effects against OGD/R-induced damage to endothelial cell barrier and function by activating PPARα/Nrf2 signaling.
