Explore the Potential Mechanism of Action and Experimental Verification of"Fenugreek Rosemary"Extract in the Treatment of Type 2 Diabetes Mellitus Based on Network Pharmacology
Objective:To analyze the potential mechanism of traditional Chinese medicine"trigonella-rosemary"in the treatment of type Ⅱ dia-betes mellitus(T2DM)by using network pharmacology related technology.Methods:The chemical components and their targets of chinese me-dicinal herbs were collected by searching the c hinese medicine database,and the targets related to type Ⅱ diabetes were searched by searching the disease target database.The network pharmacological map of active substances,active targets and disease-related targets was constructed,and the mechanism of action was explained by network analysis.Fifty SD rats were randomly divided into blank group,type 2 diabetes model group and"fenugreek rosemary"extract high-dose,medium-dose and low-dose treatment groups with 10 rats per group.The contents of to-tal triglycerides,total cholesterol,high density lipoprotein and low density lipoprotein in serum of rats were detected by the kit.The expression levels of TNF-α,IL-6,IL-2,VEGF and EGF were detected by ELISA.Results:A total of 56 active ingredients and 764 potential targets for the treatment of typeⅡdiabetes were identified from"Trigonella-rosemary".Protein interaction analysis showed that FGFR1,TNF,INSR,IL-6,MAPK14,JUN,PPARA,VEGFA,PTPN1,PTPN2,IGF1R and PTPN11 were the key targets.Animal experiments showed that the medium and high dose groups of fenugreek rosemary extract could significantly reduce the serum triglyceride and cholesterol,up-regulate the serum high density lipoprotein content,down-regulate the serum low density lipoprotein content,regulate the inflammatory factors TNF-α,IL-6,IL-2 content,and increase the expression of VEGF and EGF.Conclusion:This study systematically analyzed the mechanism of action of"trigonella-rosemary"in the treatment of type Ⅱ diabetes,and laid the foundation for the treatment of type Ⅱ diabetes with traditional chinese medicine and the research and development of new chinese medicine.
