Optimization of Extraction Process of Polysaccharide from Yunnan Huaping Cactus Gum by Response Surface Methodology and the Anticancer Activity Research
(Objective)To explore the processing parameters of the cactus gum polysaccharides and the antitumor activity.(Methods)This study optimized the extraction process of crude polysaccharides from cactus gum using single-factor experiment and response surface methodology(RSM),and evaluated the anticancer activity of the polysaccharides using the MTT assay.(Results)Firstly,the single factor experiments and response surface methodology(RSM)were used to optimize the extraction process of crude polysaccharides from cactus gum.The optimum ex-traction conditions for crude cactus gum polysaccharides by RSM is as follows:extraction time 5 hours,extraction 5 times,the liquid ratio was 1∶60(g/mL)and ethanol concentration was 70%V/V).Under this condition,the extraction rate of crude polysaccharides in cactus gum is 87.02%.Next,the optimal conditions predicted by RSM is consistent with experimental conditions,and further verification shows that the ex-traction rate is(82.38±3.19)%.Finally,the MTT assay was employed to evaluate the anti-tumor cell growth activity by the cactus gum poly-saccharide.The results showed that cactus gum polysaccharides had certain inhibitory activity on colon cancer cells(RKO)and breast cancer cells(4T1),IC50 values were 143.5 and 167.9 mg/L,respectively.(Conclusion)The extraction conditions of the cactus gum polysaccharides can effectively extract polysaccharides with high yield in this study.It was found that cactus gum polysaccharides can inhibit the growth of colon cancer cells(RKO)and breast cancer cells(4T1).
