设计合成了一种新型的硫化氢荧光探针.探针DDM是通过连接二氰基异弗尔酮和三苯胺骨架而构筑的,其结构通过核磁共振氢谱、碳谱和高分辨质谱进行表征而确定.在PBS/CH3CN缓冲溶液中(10mmol/L,pH=7.4,体积比1:1),通过紫外和荧光光谱,探究探针DDM对硫化氢的响应.结果表明,探针与H2S发生亲核加成反应,进而导致共轭体系被破坏,在397 nm处产生较强吸收,同时在490 nm处发射强荧光.探针DDM溶液在490 nm处的荧光强度与S2-呈现良好的线性关系,检出限仅为27.4 nmol/L.此外,通过生物硫醇和阴离子的选择性实验,发现探针DDM对H2S表现出特异性响应,探针DDM对H2S有较快的响应时间.
Synthesis of a Fluorescent Probe with Excellent Sensitivity for Detention Hydrogen Sulfide
In this paper,we developed a new fluorescent probe DDM for detection hydrogen sulfide(H2S).The probe was constructed by con-necting the dicyano-isophorone and trianiline skeleton and its structure were characterized by 1 H-NMR,13 C-NMR and HR-MS.In PBS/CH3CN(10 mM,pH=7.4,1/1,v/v)buffer solution,the response of probe DDM to H2S was investigated by UV and fluorescence spectra.The results indicate that the nucleophilic addition reaction between the probe and H2S leads to the destruction of the conjugated system,resul-ting in strong absorption at 397 nm and fluorescent intensity at 490 nm.The fluorescent intensity of probe DDM solution at 490 nm showed a good linear relationship with S2-,and the detection limit detection was as low as 27.4 nmol/L.In addition,the probe DDM showed a specific response to H2S through the selective experiments of biothiols and anions.Moreover,the probe DDM can detect the H2S quickly.
