HPLC法测定滇紫草及昆明滇紫草中β,β'-二甲基丙烯酰阿卡宁

(目的)建立滇紫草、昆明滇紫草中β,β'-二甲基丙烯酰阿卡宁含量的测定方法.(方法)采用安捷伦液相色谱仪,色谱柱为Thermo Acclaim 120-C18(250 mm × ø4.6 mm,5 μm),乙腈-0.2%甲酸(体积比70:30)等度洗脱,流速1.0 mL/min,柱温30 ℃,分析波长275 nm.(结果)β,β'-二甲基丙烯酰阿卡宁在24.151~301.889 ng范围内呈良好线性关系(r=0.9998),平均回收率为93.07%,RSD为2.43%(n=6);18批次样品质量分数在0.0934 mg/g~1.7584 mg/g范围内,各批样品质量分数存在差异.(结论)该方法专属性强、重复性好,符合中国药典相关要求,可用于滇紫草及其昆明滇紫草中β,β'-二甲基丙烯酰阿卡宁的含量测定.
Determination of β,β'-dimethylacrylylAkanin in Comfrey Yunnanensis and Its Related Variety Kunmingensis by HPLC
(Objective)To establish a method for the determination of β,β'-dimethylacrylylarkanin in Comfrey Yunnanensis and Comfrey Kunmin-gensis.(Methods)The chromatography was performed on a Thermo Acclaim 120-C18(250 mm × ø4.6 mm,5 μm)column with isoelution of aceto-nitrile-0.2%formic acid(70:30,V/V)at a flow rate of 1.0 mL/min,at a column temperature of 30 ℃,and at a detection wavelength of 275nm.(Results)β,β'-dimethylacrylylarkanin showed a good linear relationship in the range of 24.151-301.889 ng(r=0.9998),the average recovery was 93.07%,RSD was 2.43%(n=6).The mass fraction of 18 batches of samples was in the range of 0.0934 mg/g-1.7584 mg/g,and the mass fraction of each batch of samples was different.(Conclusion)The method is specific and reproducible,meets the relevant requirements of Chinese Pharmacopoeia.It can be used for the determination of β,β'-dimethylacrylylarkanin in C.yunnanensis and C.kunmingensis.
