Determination of β,β'-dimethylacrylylAkanin in Comfrey Yunnanensis and Its Related Variety Kunmingensis by HPLC
(Objective)To establish a method for the determination of β,β'-dimethylacrylylarkanin in Comfrey Yunnanensis and Comfrey Kunmin-gensis.(Methods)The chromatography was performed on a Thermo Acclaim 120-C18(250 mm × ø4.6 mm,5 μm)column with isoelution of aceto-nitrile-0.2％formic acid(70:30,V/V)at a flow rate of 1.0 mL/min,at a column temperature of 30 ℃,and at a detection wavelength of 275nm.(Results)β,β'-dimethylacrylylarkanin showed a good linear relationship in the range of 24.151-301.889 ng(r=0.9998),the average recovery was 93.07％,RSD was 2.43％(n=6).The mass fraction of 18 batches of samples was in the range of 0.0934 mg/g-1.7584 mg/g,and the mass fraction of each batch of samples was different.(Conclusion)The method is specific and reproducible,meets the relevant requirements of Chinese Pharmacopoeia.It can be used for the determination of β,β'-dimethylacrylylarkanin in C.yunnanensis and C.kunmingensis.
