Optimization of ultrasonic-assisted extraction technology by response surface methodology and antioxidant activity of polyphenols from Myosoton aquaticum
The aim was to improve the added value of Myosoton aquaticum resources,ultrasonic-assisted extrac-tion of polyphenols from Myosoton aquaticum was carried out and the activity of polyphenols was investigated.First-ly,the effects of ultrasonic temperature,liquid-solid ratio, ethanol concentration and ultrasonic time on the ex-traction yield of polyphenols from Myosoton aquaticum were investigated, and then the extraction process was opti-mized by Box-Behnken experimental design. Finally, the scavenging effect of polyphenols from Myosoton aquatic-um on nitrite was investigated. The results showed that the optimum extraction process parameters were as follows:ultrasonic temperature 73 ℃,liquid-solid ratio 33 mL/g,ethanol concentration 63 and ultrasonic time 41 min.Under these conditions, the average extraction rate of polyphenols from Myosoton aquaticum was 88.97 mg/g.Compared with the predicted value of the model(89.58 mg/g),the relative error was only 0.68%,which showed that the model was effective and reliable.Polyphenols from Myosoton aquaticum had a certain nitrite scavenging a-bility,and its IC50 value was 1.82 mg/L,and there was a dose-effect relationship with nitrite scavenging rate. This study provided a new way for the comprehensive development and utilization of Myosoton aquaticum.
