微电泳吗啡和纳洛酮对大鼠腹侧苍白球神经元自发放电活动的影响

目的 通过微电泳法探讨吗啡和纳洛酮对大鼠腹侧苍白球(VP)神经元自发放电活动的影响.方法 采用微电泳的方法,对30只大鼠VP神经元进行微电泳吗啡和纳洛酮,观察并记录VP神经元的自发放电情况.结果 吗啡增加VP神经元的放电频率,纳洛酮降低VP神经元的放电频率(P<0.05).在微电泳吗啡的基础上进行130 Hz高频电刺激伏隔核(NAc),大部分神经元出现抑制反应.结论 吗啡增加VP神经元的放电频率,纳洛酮降低VP神经元的放电频率;在微电泳吗啡的基础上高频电刺激NAC,大多数VP神经元出现抑制反应,可为临床治疗药物成瘾提供实验依据.
Abstract
Objective To investigate the effects of morphine and naloxone on the spontaneous firing activity of ventral globus pallidus(VP)neurons in rats by microelectrophoresis.Methods Microelectrophoresis of morphine and naloxone was performed on VP neurons of 30 rats,and the spontaneous discharge of VP neurons was observed and recorded.Results Morphine increased the discharge frequency of VP neurons,and naloxone decreased the discharge frequency of VP neurons(P<0.05).The nucleus accumbens(NAc)was stimulated by 130 Hz high-frequency electrical stimulation based on microelectrophoretic morphine,and most neurons showed inhibitory response.Conclusion Morphine increased the discharge frequency of VP neurons,and naloxone decreased the discharge frequency of VP neurons.High frequency electrical stimulation of NAc was performed on the basis of morphine microelectrophoresis,and most VP neurons showed inhibition response,which could provide experimental basis for clinical treatment of drug addiction.
