VEGF和E-选择素在牙周炎伴动脉粥样硬化大鼠血清中的变化

摘要
目的:检测牙周炎和动脉粥样硬化大鼠血清中血管内皮生长因子(VEGF)及E-选择素(E-selectin)的水平.方法:选取48只6月龄雄性Wistar大鼠,随机分为4组(n=12):正常对照组(A组)、牙周炎组(B组)、动脉粥样硬化组(C组);牙周炎合并动脉粥样硬化组(D组),分别进行相应的建模处理后,观察各组大鼠牙周组织和颈总动脉血管的病理改变;酶联免疫吸附法(ELISA)检测血清中VEGF、E-选择素的水平.结果:牙周病理组织学观察显示,B、D组大鼠牙周炎症较A、C组明显;颈总动脉病理组织观察显示,C、D组形成粥样硬化病变.血清中VEGF、E-选择素水平均以D组最高,由高到低依次为D组＞C组＞B组＞A组(P＜0.05).结论:VEGF、E-选择素对牙周炎、动脉粥样硬化具有促进作用,且牙周炎和动脉粥样硬化可能存在相互影响的关系.
Abstract
AIM:To observe the expression of vascular endothelial growth factor (VEGF) and E-selectin in serum of rats with experimental periodontitis and atherosclerosis.METHODS:48 male Wistar rats (6-month old) were randomly divided into group A (control group),group B (periodontitis group),group C (arherosclerosis group) and group D (periodontitis plus atherosclerosis)(n =12).At the end of 4th W,2 rats were sacrificed to confirm the successful establishment of periodontitis and arherosclerosis.The remaining rats were sacrificed at the end of 12 W.Maxilla were taken for HE staining,blood were taken for the detection of VEGF and E-selectin expression by ELISA.RESUITS:Histological observation showed that periodontitic changes in group B and D.Atherosclerotic lesions could be observed in carotid artery of group C and D.Serum VEGF and E-selectin level was higher in groups B,C and D than that in group A (P ＜0.05).Moreover,serum VEGF and E-selectin level in group D was higher than that in group B and C (P ＜ 0.05).CONCLUSION:Periodontitis and atherosclerosis have a positive correlation.
